Identification of The Expression Level of HSP27, HSP90 Genes in the Oviduct Epithelial Cells of Iraqi Cows with Endometritis

Abstract

The present study was carried out to study the HSP27 and HSP90 genes'. Expression in Oviduct epithelial cells of Iraqi cows with endometritis in comparison with it in the  oviduct epithelial cells of healthy Iraqi cows. Uterine samples (N=50)  were collected from cows with macroscopic changes in the uterus. The collection samples were done twice a weekly for four months, from October 2024 to February 2025.The samples were taken from the uterine endometrium and oviduct ampulla for molecular and histopathology studies. The histological examination endometrial samples (N=30) of the cows' reproductive tracts exhibited histological, while the other samples (N=20) were normal. The molecular study involved RNA extraction from oviduct tissue ampulla and RNA concentration measurement by Quantus™ Fluorometer , cDNA syntheses and Quantitative Reverse transcriptase PCR (RT-qPCR). Preparation. The gene expression analysis of HSP90 using RT-qPCR technology revealed a significant up-regulation in cows oviduct ampulla within endometritis compared to normal endometrium cows (P < 0.0001) and significantly different (P < 0.01). The expression level in oviduct cows with endometritis (1.630 fold increase) compared to the oviduct with normal endometrium cows (1.026) , The gene expression analysis of HSP27. Using RT-qPCR technology revealed a significant up-regulation in cows oviduct within endometritis compared to healthy cows (P< 0.0001) and significantly different (P < 0.01) the expression level in cows oviduct with endometritis (1.675 fold)  in comparison to that in the oviduct with normal endometrium (1.043) . In conclusion, endometritis associated increased expression of HSP90 and HSP27. The presence of gross lesion does not be necessarily indicate microscopic pathological change in endometrium.
